西洋参果总皂苷对冠状动脉结扎犬血流动力学及心肌缺血的影响

Effect of panax quinquefolium fruit saponin on hemodynamics and acute myocardial ischemia in coronary artery ligated dogs

目的:观察西洋参果总皂苷(PQFS)对实验性心肌缺血犬的血流动力学及其相关指标的影响。方法:将实验犬随机分为对照组、银杏叶片组(24 mg.kg-1)及PQFS 24、12和6 mg.kg-13个剂量组,每组6只。结扎犬左冠状动脉前降支(LAD)造成急性心肌缺血模型,测定血流动力学指标、心肌梗塞面积及血清酶学变化。结果:与对照组比较,PQFS 24、12和6 mg.kg-1组冠状动脉血流量增加(P<0.05),心肌耗氧量减少(P<0.05),心肌梗塞面积缩小(P<0.01),血清中谷草转氨酶(AST)、肌酸激酶(CK)、乳酸脱氢酶(LDH-L)活性降低(P<0.01,P<0.05),血清中丙二醛(MDA)含量减少(P<0.01,P<0.05),超氧化物歧化酶(SOD)活性增加(P<0.01,P<0.05)。结论:PQFS可降低冠脉阻力,增加冠脉流量,减少心肌耗氧量,具有保护缺血心肌作用。

Objective To observe the effect of panax quinquefolium fruit saponin （PQFS） on hemodynamics and correlative indexes in the coronary artery ligated dogs. Methods The dogs were divided into five groups randomly （n=6）： control group, gingkgo leaf tablet group （24 mg. kg^-1）, 24, 12 and 6 mg·kg^-1 PQFS groups. The myocardial infarction size （MIS） and the changes of the serum enzymes were determined by using the acute myocardial infarction models with the ligation of left anterior descending coronary artery of dogs. The parameters of hemodynamics and myocardial oxygen metabolism were measured in the anesthetic dogs with thoracotomy. Results Compared with control group, the myocardial blood flow was increased （P〈0.05）, the coronary resistance and myocardial oxygen were decreased （P〈0.05） in dogs treated with PQFS of 24,12 and 6 mg·kg^-1 The MIS and the activities of serum glutamic oxaloacetict ransaminase enzyme （AST）,creatine kinse （CK） and lactic debydrogenase （LDH-L） were decreased （P〈0.01, P〈0.05） in acute myocardial infarction dogs treated with PQFS of 24, 12 and 6 mg·kg^-1 The activities of superoxide dismutase （SOD） was increased （P〈0.01, P〈20.05）, but the content of serum malonyl dialdebyde （MDA） was decreased （P〈0.01, P〈0.05）. Conclusion PQFS has protective effect on myocardial iscbemia through decreasing myocardial oxygen consumption and increasing myocardial blood flow in dogs.
