摘要
目的探讨雌激素在单侧输尿管梗阻(UUO)大鼠肾间质纤维化模型中对肾小管上皮细胞表型转化的影响。方法雌性SD大鼠30只,随机分为4组:对照组;生理雌激素组(单纯UUO);低雌激素组(UUO+OVX);高雌激素组(UUO+E2)。21d后处死各组大鼠,用光镜观察梗阻肾组织病理变化,并用免疫组化和RT-PCR方法检测肾组织α-SMA蛋白和mRNA的表达。结果生理雌激素组出现肾小管间质纤维化改变,其肾组织α-SMA表达上调,与对照组相比有统计学意义(P<0.01);和生理雌激素组相比,低雌激素组纤维化病变加重,α-SMA表达明显增多(P<0.05);而高雌激素组则病变减轻,上述表达减少(均为P<0.05)。结论雌激素可能通过抑制UUO大鼠肾小管上皮细胞表型转化而发挥延缓肾间质纤维化的作用。
[Objective] To evaluate the effect of estrogen on tubular epithelial cell phenotype transdifferentiation of tubulointerstitial fibrosis in unilateral ureteral obstruction (UUO) rats. [Methdos] Thirty Sprague-Dawley femal rats were randomly divided into four groups: control group; normal estrogen group (rats only underwent UUO); low estrogen group (rats were ovariectomized and underwent UUO); high estrogen group (rats underwent UUO and received 17β-estradiol injection). The rats were killed after 21 days, renal tissues were examined by microscope. Immunohistochemistry and reverse transcfiption-polymerase chain reaction were applied to determine the protein and the mRNA expression of α-SMA. [Result] Compared with control group, there appeared tubulointerstitial fibrosis in normal estrogen group, the expression of α-SMA remarkably increased (P 〈0.01). Compared with normal estrogen group, the lesion was remarkably aggravated in low estrogen group, the expression of α-SMA enhanced (P 〈0.05), and in high estrogen group the lesion was attenuated, the expression of α-SMA decreased (P 〈0.05). [Conclusion] Estrogen can prevent the development of interstitial fibrosis ,the mechanism may with it restrain the tubular epithelial cell phenotype transdifferentiation in UUO rats.
