摘要
优化了黄豆芽愈伤组织培养条件,建立了合适的黄豆芽细胞悬浮培养体系。以在25℃无激素条件下自然生长的豆芽子叶、叶芽、芽茎、芽根等部位为外植体诱导愈伤组织,以次生代谢产物Vc积累最多为目标,考察外植体、基础培养基、萘乙酸(NAA)浓度以及pH值的影响。结果表明:叶芽在pH值为5.0的B5培养基中,能有效诱导愈伤组织,且目标产物Vc积累最多。低浓度NAA对黄豆芽愈伤组织诱导及次生代谢产物Vc积累影响不大,随浓度增加,影响加大。
The culture condition of soybean sprout callus were optimized and the system of suspension of soybean sprout cell was set up, Under the condition of hormone-frce at 25 ℃,the callus were inducted with the colyledon, leaf bud, bud stem and bud root firm bean sprout which grew naturally. Based on the most accumulation of secondary metaholites Vitamin C as an object, the effect of explants, basal medium, concentration of NAA and pH on Vc accumulation were compared. Tire result showed that the callus could be effectively induced from the leaf bud cell of soybean sprout cultured in B5 medium at pH 5.0 and the production of Vc was most. NAA in lower concentration little affected the induction of the callus of soybean sprout and the accumulation of the secondary metabolites Vitamin C, but the effect was increased as the increasing of NAA concentration.
