摘要
采用单因素试验和正交试验,考察了Mg2+、Ca2+、Fe2+、Mn2+和Zn2+5种金属离子对悬浮培养霍山石斛类原球茎合成活性多糖的影响。结果表明:1.5 mmol/L Mg2+4、.5 mmol/L Ca2+0、.1 mmol/L Fe2+0、.5 mmol/L Mn2+和0.06 mmol/L Zn2+的组合最有利于类原球茎多糖的合成,在此组合下多糖产量达最高(5.87 g/L),比优化前提高2.49倍,比标准MS培养基提高31倍,且5种离子对类原球茎合成多糖影响的主次顺序为Mg2+>Ca2+>Mn2+>Fe2+>Zn2+。
This research is concerned with the effect of Mg^2+ , Ca^2+ , Fe^2+ , Mn^2+ and Zn^2+ on polysaccharide synthesis in Pretoconn-like body of Dendrobium husoshanense in suspension culture through the orthogonal matrix method with one factor-at-a-time. The results showed that the optimal combination for polysaccharide accumulation in protocorm-like body was 1.5 mmol/L Mg^2+ ,4.5 mmol/L Ca^2+ ,0.1 mmol/L Fe^2+ ,0.5 mmol/L Mn^2+ and 0.06 mmol/L Zn^2+. The peak of the polysaccharide production(5.87 g/L) was obtained in the culture medium with the same ion combination mentioned above, which was increased 2.49-fold than that of the former experiment and was increased 31-fold than that obtained in standard MS. Furthermore, the responsible order of metal ion in the polysaccharide synthesis was Mg^2+ 〉 Ca^2+ 〉 Mn^2+ 〉 Fe^2+ 〉 Zn^2+.
