摘要 目的 研究同型半胱氨酸 (Hcy)对大鼠主动脉平滑肌细胞增殖及增殖细胞核抗原基因(cyclinD1)mRNA表达的影响。方法 采用同位素技术测定3H TdR参入法和核酸分子杂交技术检测大鼠主动脉平滑肌细胞增殖及增殖细胞核抗原基因 (cyclinD1)mRNA表达的水平。结果 Hcy能促进VSMC增殖 ,并且具有剂量效应关系 ,同时可增加cyclinD1mRNA表达水平 3 2倍。结论 Hcy促进VSMC增殖可能是通过增加cyclinD1mRNA表达水平的途径实现的 ,使静止期细胞进入细胞分裂周期 ,从而促进VSMC迅速增殖。 Objective To study the effect of homocysteine (Hcy)on the proliferation of vascular smooth muscle ceils (VSMC)and the expression of cyclin D1 mRNA. Methods 3^H-TdR incorporation and Northern blot were assayed to detect the amount of 3^H-TdR and the levels of cyclin D1mRNA expression. Results Hcy induced the proliferation of VSMC and there was a dose-dependent relationship in it. Hcy increased the expression of cyclin D1 mRNA in rat aortic smooth muscle cells by 3.2-fold. Conclusion Hcy induced the proliferation of VSMC probably by the increased the expression of cyclin D1mRNA, indicating the Hcy induced the mRNA of cyclin is important for the reentry of quiescent VSMC into the cell cycle.
