摘要
摸索出一种新的DNA提取方法——CTAB改良法,从梨成熟叶片中提取到高质量、高纯度的DNA。同时,分析了模板DNA和dNTPs浓度、TaqDNA聚合酶用量等对RAPD反应体系的影响,建立了一套适合梨基因组RAPD分析的技术体系。
A new DNA extraction method -- improved CTAB method was found which could extracted high-quality and high-purity DNA from ripe leaves of pears. Meanwhile,some factors were optimized,such as the concentration of DNA as template,dNTPs and usage of Taq DNA polymerase.A suitable reaction system for RAPD-PCR of pear genome was established.
