玉米紫色植株花色苷色素延缓衰老的功能 被引量:15

摘要 目的:通过观察玉米紫色植株色素对果蝇寿命、体内超氧化物歧化酶活性和丙二醛含量的影响,分析玉米紫色植株色素延缓衰老的作用。方法:实验于2004-12/2005-12于沈阳医学院营养与食品卫生学教研室实验室完成。收集10h内羽化未交配的黑腹果蝇6400只,雌雄各半。①果蝇寿命实验:将1600只果蝇按体质量大小随机分为4组,即对照组、含0.2%,1.0%,5.0%玉米紫色植株色素组,每组雌雄果蝇各200只,雌雄分开饲养。对照组给予普通玉米培养基,含0.2%,1.0%,5.0%玉米紫色植株色素组分别给予含玉米紫色植株色素0.2%,1.0%,5.0%的玉米培养基喂饲,每4d更新1次培养基。每天定时记录果蝇死亡数,直至果蝇全部死亡。计算果蝇平均寿命、半数死亡期和最高寿命穴每组最后死亡的20只果蝇寿命的算数平均数雪。②抗氧化实验:选择果蝇4800只,分组和喂养方法同寿命试验,每组雌雄果蝇各600只。将实验第40天雄性果蝇和第54天雌性果蝇每50mg为一个样本,匀浆,测定超氧化物歧化酶活性和丙二醛含量。结果:进入结果分析果蝇数量为6353只,在实验过程中果蝇飞失。①与对照组比较,含0.2%,1.0%,5.0%玉米紫色植株色素组雌雄果蝇寿命均明显延长,含1.0%玉米紫色植株色素组作用明显,其中雌雄果蝇平均寿命分别延长8.7%和12.0%,最高寿命分别延长15.5%和6.2%。各组之间半数死亡天数差异无显著性意义(P>0.05)。②含1.0%玉米紫色植株色素组雌性果蝇体内超氧化物歧化酶活性显著高于对照组和含5.0%玉米紫色植株色素组眼(48.976±5.651,37.674±11.819,41.025±7.902)μkat/g(P<0.01)演,丙二醛含量显著低于对照组和含5.0%玉米紫色植株色素组眼(34.9±5.2,40.9±6.3,40.6±5.9)nmol/g(P<0.01)演。含1.0%玉米紫色植株色素组雄性果蝇体内超氧化物歧化酶活性显著高于对照组眼(50.743±7.602,44.058±6.551)μkat/g(P<0.05)演,而丙二醛含量显著低于对照组眼(34.9±5.2,40.9±6.3)nmol/g(P<0.05)演。结论:玉米紫色植株色素能延长果蝇寿命,提高果蝇体内超氧化物歧化酶活性,降低丙二醛含量,其中含1.0%玉米紫色植株色素组表现最为显著。其延缓衰老的作用可能是通过提高抗氧化和促进自由基清除的能力而产生的。 AIM: To explore the effect of maize purple plant pigment on delay of senility by observing its influence on drosophila's lifespan, activity of superoxifde dismutase (SOD) and content of malondialdehyde (MDA). METHODS: The experiment was conducted in the laboratory of Teaching and Research Section of Nutrition and Food Hygiene, Shenyang Medical College between December 2004 and December 2005. Totally 6 400 unmated drosophila melanogasters, female and male in half, eclosion in 10 hours were selected. ① Lifespan of the drosophila: 1 600 unmated drosophilae melanogasters were randomly divided into four groups: control group and three-dosage of maize purple plant pigment experiment groups according to body mass. The control group was given normal maize medium; the experiment groups were fed with culture medium contained 0.2%, 1.0% and 5.0% maize purple plant pigment, respectively, and the medium was renewed every 4 days. The death number of drosophila melanogasters was recorded until all died. Meanwhile, mean lifespar,, half died time and highest lifespan (mean of the last died 20 ones in each group) were calculated, respectively. ②Experiment of antioxidation: 4 800 unmated drosophilae melanogasters were randomly divided into groups and were fed the same as the experiment of the drosophila's lifespan, The samples of male drosophila melanogasters at the 40^th day and female ones at the 54^th day (every 50 mg as one sample) were taken to determine the activity of SOD and content of MDA, respectively. RESULTS: Totally 6 353 drnsophila melanogasters were involved in the result analysis with some flying out. ①Compared with the control group, the lifespan of drosophila melanogasters in experiment groups were prolonged by maize purple plant pigment, especially the experiment group containing 1.0% maize purple plant pigment, in which the mean lifespan, highest lifespan of both female and male drosophila melanogasters were prolonged by 8.7% and 12,0%, 15.5% and 6.2%, respectively, There was no significant difference in half died days among groups. ② In the experiment group containing 1.0% maize purple plant pigment, the SOD activity in female drosophila melanogasters was significantly higher than that in the control group and 5.0% group [(48.976±5.651, 37.674± 11.819, 41.025±7.902) μkat/g, P 〈 0.01], while the MDA content was obviously lower than that in control group and the 5.0% group [(34.9±5.2, 40.9±6.3, 40.6±5.9) nmol/g, (P 〈 0.01)]. The SOD activity in male drosophila melanogasters of 1.0% group was markedly higher than that in the control group [(50.743 ±7.602, 44.058±6.551) μkat/g, P〈 0.05], wbile the MDA content was obviously lower than that in control group [(34.9±5.2, 40.9±6.3) nmol/g, (P 〈 0.05)]. CONCLUSON: Maize purple plant pigment can prolong lifespan, increase activity of SOD and decrease MDA coutent in drosophila melanogasters, especially with dose of 1.0%. It may delay aging by increasing alltioxidation and removing free radical.
