玉竹多糖对衰老模型鼠细胞及体液免疫功能的影响 被引量:14

摘要 目的:观察玉竹多糖对D-半乳糖致亚急性衰老模型小鼠免疫功能的影响,分析玉竹多糖抗衰老的可能作用途径。方法:实验于2004-07/2005-05在锦州医学院免疫实验室完成。取昆明小鼠50只,按随机数字表法分为5组,即正常对照组、衰老模型组和0.5g/kg,1g/kg,2g/kg玉竹多糖组。除正常对照组外,其余各组每只颈背部皮下注射50g/LD-半乳糖0.5mL,连续6周。同时0.5g/kg,1g/kg,2g/kg玉竹多糖组分别腹腔注射0.5g/kg,1g/kg,2g/kg,0.5mL的玉竹多糖给予治疗,正常对照组和衰老模型组腹腔注射生理盐水0.5mL/只。6周后,将小鼠称重并麻醉剖取小鼠胸腺和脾脏,计算脾指数穴mg/g雪=脾质量穴mg雪/小鼠体质量穴g雪,同法计算胸腺指数。应用MTT法测定脾淋巴细胞转化指数。用免疫荧光法测定脾T淋巴细胞亚群细胞数,观察其对免疫功能的影响。结果:50只小鼠全部进入结果分析,无脱失。①与正常对照组比较,衰老模型组小鼠胸腺指数和脾指数显著下降(P<0.05~0.01);脾T,B淋巴细胞转化刺激指数显著降低(P<0.05~0.01);CD8+细胞数减少穴P<0.01雪,CD4+/CD8+比值增加穴P<0.01雪。②与衰老模型组比较,0.5g/kg,1g/kg,2g/kg玉竹多糖组小鼠的脾指数、胸腺指数、B淋巴细胞转化指数及CD8+细胞数均显著提高(P<0.05~0.01),而CD4+/CD8+比值显著降低穴P<0.01雪,但3个剂量组之间差异无显著性意义穴P>0.05雪。结论:玉竹多糖可明显改善D-半乳糖致衰老模型小鼠的细胞及体液免疫功能,提高胸腺指数和脾指数,增加脾T,B淋巴细胞转化刺激指数和CD8+细胞的数量,恢复CD4+/CD8+比值的失衡状态,从而延缓机体的免疫衰老。玉竹多糖不同剂量组间无明显的剂量依赖性。 AIM: To observe the effect of polygonatum odoratum polysaceharide on the immunological function of aging model mice induced by D-galactose, and analyze the possible mechanism of anti-aging effect of polygonatum odoratum polysaccharide. METHODS: The experiment was completed in the Immunological Labo- ratory of Jinzhou Medical University between July 2004 and May 2005. Fifty Kunming mice were randomly divided into five groups: normal control group, senile model group and three test groups of 0.5 g/kg, 1 g/kg and 2 g/kg polygonatum odoratum polysaccharide, Except the normal control group, the mice of other groups were given the subcutaneous injection of 0.5 mL D-galactose (50 g/L) for 6 weeks. Meanwhile, the test mice were injected intraperitoneally 0.5 mL polygonatum odoratum polysaccharide while mice of normal control group and senile model group were treated with 0.5 mL saline. Six weeks later, all the mice were weighted and anesthetized to fetch thymus and spleen for measuring thymus index and spleen index [Index(mg/g)=Mass(mg)/Body mass of mice(g)]. Spleen lymphocyte transforming index was measured with MTF method, and subtypes of T lymphocyte were counted by immunofluorescence to observe the influence on the immunological function. RESULTS: All the 50 mice were put into the result analysis without drop. ①Compare the normal control group, the thymus index, spleen index as well as spleen T and B lymphocyte transforming index of mice remarkably decreased (P〈 0.05-0.01); The number of CD8^+ T lymphocyte reduced while the ratio of CD4^+/CD8^+ increased in the senile model group (P 〈 0.01). ②In contrast with senile model group, three dosages of polygonaturn odoratum polysaccharide increased the thymus index, spleen index, spleen B lymphocyte transforming index and the number of CD8^+ T lymphocyte (P 〈 0.05- 0.01), but reduced the ratio of CD4^+/CD8^+ notably (P 〈 0.01). There was no significant difference in the three groups of polygonatum odoratum polysaccharide (P 〉 0.05). CONCLUSION: Polygonatum odoratum polysaccharide can improve cellular immunity and humoral immunity of the aging model mice induced by D-galactose, elevate the thymus index and spleen index, heighten the spleen T and B lymphocyte transforming index and the number of CD8^+ T lymphocyte, and recovery the balance of CD4^+/CD8^+ so as to delay senile immunity. There is no obvious dosage-dependence among the groups of different dose of polygonatum odoratum polysaccharide.
