大鼠慢性阻塞性肺病模型体内TNF-α和VEGF含量的变化及其意义 被引量:7

摘要 目的探讨肿瘤坏死因子-α(TNF-α)及血管内皮生长因子(VEGF)在慢性阻塞性肺病(COPD)发病中的作用。方法香烟熏吸法复制COPD大鼠模型。将20只清洁级Wister大鼠随机分为对照组和COPD模型组,每组各10只。肺组织切片行苏木精-伊红染色,测定两组大鼠血清、肺组织匀浆及支气管肺泡灌洗液(BALF)中TNF-α和VEGF的含量,并用RT-PCR法测定肺组织中VEGF mRNA的表达。结果模型组大鼠血清、肺组织匀浆及BALF中TNF-α含量较对照组明显升高(P<0.01),而VEGF的含量较对照组有明显降低(P<0.01,P<0.05)。模型组大鼠肺组织内VEGF的表达较对照组有明显下降(P<0.05)。结论TNF-α和VEGF参与了COPD的发病过程且在COPD细胞凋亡过程中起重要作用。 Purpose To discuss the fuction of TNF-α and VEGF on the experimental rat model with chronic obstructive pulmonary disease. Methods The rat model with COPD was established by exposure with cigarette smoke (4 hours per day,5 days per week, up to 16 weeks). The randomized-controlled-trails was used in the experiment. Twenty male Wister rats were divided randomly into two groups: the normal control group and the model group, 10 rats each. Histopathological changes were observed. The slides of lung tissue were dyeing with Hematoxylin-Eosin. The TNF-α and VEGF contents in the blood serum, lung tissue and BALF of each group were measured by Elisa. The expression of VEGF mRNA was measured by reverse transcription-poiymerase chain reaction (RT-PCR). Results The TNF-α contents in the blood serum, lung tissue and BALF of the model group were significantly improved than that of the normal group (P〈0.01) ,while VEGF contents reduced (P〈 0.01 or P〈 0. 05 ). The expression of VEGF mRNA of the model group was lower than that of the normal control group (P〈0.05). Conclusions TNF-α and VEGF play an important role in the apoptosis of COPD.
