摘要
目的探讨大鼠骨髓基质细胞(MSCs)体外诱导分化为神经细胞的可行性,为MSCs移植治疗缺血性脑血管病提供实验依据。方法从大鼠骨髓中分离培养骨髓基质细胞,应用bFGF进行预诱导,然后用ATRA和NGF来诱导骨髓基质细胞,观察细胞的形态变化,用免疫组化法检测细胞的NSE和GFAP的表达。结果大鼠MSCs经ATRA和NGF诱导后,细胞的形态发生改变,免疫组化显示部分细胞表达NSE和GFAP。结论骨髓基质细胞可被ATRA和NGF在体外诱导分化为神经细胞和胶质细胞。
Objective To study the feasibility of in vitro inducing different of rat bone marrow stromal ceils (MSCs) into neurons, and to offer reference for transplantation MSCs for the theaphy of ischemic cerebrovascular disease. Methods The bone morrow stromal cells from rats were isolated. MSCs were incubated with bFGF for 24 hours, then the media were replaced by ATRA and NGF. The morphological changes of the cells were observed phase contrast microscope. The morphological changes of induced MSCs were observed, we monitored expression of NSE and GFAP by indirect immunocytochemisty staining. Results After an hour of induction with ATRA and NGF, some cells showed morphological changes, and stained positively with NSE and GFAP. Conclusions ATRA and NGF can induce MSCs into neurons and glia cells in vitro.
