摘要
目的观察葛根素提取液对豚鼠乳头肌动作电位及延迟整流钾电流的影响。方法用常规微电极方法记录豚鼠乳头肌动作电位,用全细胞膜片钳技术记录游离心室肌细胞钾离子流。结果①不同浓度的葛根素均能延长豚鼠乳头肌细胞动作电位时程(APD)。②不同浓度的葛根素均能抑制延迟整流钾电流。结论葛根素明显延长APD,抑制延迟整流钾电流,具有明显的浓度依赖关系,可能是其抗心律失常的电生理机制。
OBJECTIVE To study the effect of puerarin on action potential and delayed rectifier potassium current in guinea pig papillary muscles, METHODS Action potentical was recorded by the standard microelectrode technique, Potassium current of the cardiac muscle was recorded by whole-cell variant patch-clamp technique,RESULTS ① All the different concentration puerarin prolonged the action potential duration,②All the different concentration puerarin inhibited delayed rectifier potassium current. CONCLUSION The puerarin can prolong APD and inhibit delayed rectifier potassium current in an obvious concentration-dependent manner, which may be one of critical basis on antiarrhythmin.
