p27和p21表达与结肠癌细胞浸润转移的组织微阵列研究

Study on the Expression of p27,p21 and the Invasion and Metastasis in Colon Cancer

目的:研究肿瘤相关基因p27、p21在结肠癌组织及其转移灶中的表达及其与患者预后的关系。方法:收集随访资料完整并同时具有淋巴结转移的结肠癌标本100例,按预先设计的需要制作成组织芯片,进行p27、p21免疫组化染色。根据染色指数(stainingindex,阳性细胞百分数×染色强度)观察分析结肠癌原发灶与转移灶的关系,并行相关分析。结果:100例结肠癌标本中,转移灶组织中p27、p21的表达均高于原发灶肿瘤组织的表达,差别均具有统计学意义(P<0.01)。p27、p21的表达与患者的预后呈负相关。结论:肿瘤相关基因p27、p21在已转移的结肠癌组织中的高表达和表达转移可能预示患者预后更差。

Objective： To investigate the expression of suppressor gene p27, p21 in primary and metastasis colon cancer and the relationship between p27, p21 protein expression and prognosis. Methods： One hundred cases of colon cancer with lymph node metastasis were collected in our study. Tissue microarray was prepared according to previous design. Immunohistochemical staining for p27, p21 was performed using SP technique. The primary and metastasis of colon cancer were analyzed according to staining index. Results： The expression of p27 and p21 in metastasis focus was significantly higher than that in primary tumor （P 〈 0.01）. The intensity of p27, p21 expression had negative correlation with patient＇s prognosis （P 〈 0.05）. Conclusion： Higher expression and expression shift of p27, p21 in some metastasis colon cancer may predict poorer prognosis.
