摘要
目的探讨氯化镧对人脑胶质瘤细胞株SHG 44细胞诱导凋亡及其作用机制。方法体外培养人胶质瘤细胞,流式细胞仪、电镜检查细胞周期和超微结构的改变,原位杂交技术及免疫组化染色技术检测bcl-2、bax基因、蛋白表达情况。结果氯化镧能促进入脑胶质瘤SHG 44细胞发生凋亡,明显增加G1期DNA含量,减少S期DNA含量。使bax基因、蛋白表达显著增强,bcl-2、bax基因蛋白表达显著减弱(P<0.05)。结论氯化镧可抑制人脑胶质瘤SHG 44细胞的生长,并促进其凋亡。
Objective: To investigate the feasibility and mechanism of LaCl3 induced apoptosis in human glioma SHG44. Methods: The human glioma SHG44 cells were cultured in vitro,the cell circle and ultrastructure were detected by flow cytometry and electron microscope. The protein expression and translation of bax,bcl-2 were detected by immunohistochemistry and in situ hybridization methods. Results : The LaCl3 could inhibit gene the growth of SHG44 and induce apoptosis. The DNA content of SHG44 increased in G1 cycle and decreased in S cycle obviously. Typical apoptosis cells appeared. Both of protein expression and mRNA translation of bcl-2 gene decreased, the bax gene protein expression and mRNA translation increased( P 〈 0. 05). Conclusion: LaCl3 can inhibit the proliferation of SHG44 cells in vitro and cause apoptosis.
