重组小鼠pcDNA3.1(+)/GDF-5真核表达质粒的构建及其意义 被引量:1

摘要 根据基因库中小鼠生长分化因子-5(GDF-5)序列设计并合成引物,提取孕14 d小鼠胚胎肢芽组织总RNA,行RT-PCR扩增,将扩增产物GDF-5基因片断插入至pcDNA 3.1(+)载体并转化大肠杆菌DH 5α,提取重组质粒,酶切鉴定及测序。结果:DNA琼脂糖凝胶电泳和双酶切均显示出一长约1 603 bp的带;测序结果与基因库中的序列完全相同。认为重组小鼠pcDNA 3.1(+)/GDF-5真核表达质粒构建成功,为进一步研究其在软骨发育中的作用奠定了基础。
