摘要
目的:观察中药天麻钩藤饮对愤怒应激大鼠下丘脑单胺类递质含量的影响。方法:本实验参照Megan E等方法建立一种更接近人类发怒的愤怒动物模型,采用高效液相色谱———电化学法检测大鼠下丘脑单胺类递质去甲肾上腺素(NE)、肾上腺素(E)、3-甲氧基-4-羟基苯二醇(MHPG)、多巴胺(DA)、3,4-二羟基苯乙酸(DOPAC)、5-羟吲哚乙酸(5-HIAA)、5-羟色胺(5-HT)和高香草酸(HVA)的含量。结果:与正常对照组比较,模型组大鼠下丘脑NE、DA的含量升高(P<0.05),代谢产物DOPAC、5-HIAA的含量也明显升高(P<0.01);而E、5-HT、MHPG、HAV的含量变化不显著(P>0.05)。与模型组比较,给药组大鼠下丘脑NE、DA及代谢产物DOPAC、5-HIAA的含量明显降低(P<0.05),5-HT的含量升高(P<0.01),E、MHPG、HVA的含量则无显著差异。结论:愤怒刺激可使大鼠下丘脑内单胺类神经递质NE、DA及其代谢产物DOPAC、5-HIAA的含量升高;天麻钩藤饮对大鼠受愤怒应激后所致下丘脑单胺类神经递质及其代谢产物的紊乱具有调节作用。
Objective: To investigate the changes of monoamine in hypothalamus of the model rats with angry stress and the effects of TianMaGouTeng Decoction (TMGTD) . Methods: To establish the angry stress rats model according to Megan's method and determine the contents of monoamine neurotransmitter and their metabolite by HPLC-ECD. Results: Compared with normal group. The contents of NE, DA and DOPAC, 5-HIAA have rose singnificantly in model rats ( P 〈 0.05 ). but those of E, 5-HT, MHPG, HAV have no significant discrepancy ( P 〉 0.05) . The lower contents of NE, DA, DOPAC and 5-HIAA and had been observed in TMGTD group. On the contrary, the contents of those rose signifieantly in model group. Compared with model group, TMGTD ean inerease the contents 5-HT effeetively( P 〈 0.01 ) .The contents of E, MHPG and HVA have no signifieant diserepaney ( P 〉 0.05 ) .Conelusions : anger stress can increase the contents of NE, DA, DOPAC and 5-HIAA. TMGTD can regulate the abnormal eontents of monoamine neurotransmitter and their metabolite in hypothalamus of rats with anger stress.
