摘要
目的:探讨维生素C对甘露醇引起血管损伤及其诱导的血管内皮细胞凋亡基因表达的影响。方法:(1)动物实验:将健康大白兔随机分4组,A组在兔耳缘静脉注射甘露醇;B、C、D组分别以生理盐水、维生素C、维生素E预处理后再静脉推注甘露醇,电镜、光镜下观察局部血管的损伤。(2)利用Biostar H-40s型芯片,检测甘露醇分别在应用三种保护剂前后对凋亡基因表达的影响。结果:(1)动物实验:血管损伤程度上,A组为重度;B组为中~重度;D组为轻度;C组未见明显病理改变。(2)基因芯片检测:330条凋亡基因中,生理盐水组芯片无改变;维生素C组中45条促凋亡基因表达下调,34条抗凋亡基因表达上调;维生素E组3条促凋亡基因表达下调。结论:维生素C通过抑制血管内皮细胞凋亡而起到抑制甘露醇血管损伤的作用。
Objective:To research the gene expressions of injury on blood vessel and vascular endothelial cell apoptosis induced by mannltol in using vitamin C.Methods: (1) Animal experiment: healthy rabbits were randomized into 4 groups. Group A was injected with mannltol into the vein of the rabbit ear. Group B, group C,and group D, were injected first with NS, VitC and VitE respectively to pretreat the vessel, and then with mannitol. Electronic lens and optical microscope were used to observe the injury of blood vessel. (2) Arrays of type Biostar H-40s were used to test the changes of apoptosis gene expressions induced by marmitol before and after using protective agents.Results: (1) As for the degree of injury of the blood vessel, group A was serious, group B was moderate to serious, and group D is slight. No pathological changes were observed in Group C. (2) All 330 apoptesis-related genes: in chips pretreated by normal saline, none was significantly different. In chips pretreated by vitamin C, 45 apoptosis genes were down-regulated, 34 anti-apoptosis genes were up-regulated. In chips pretreated by vitamin E, only 3 apoptosis genes were down-regulated. Conclusions:Vitamln C can protect vascular endothelial cell.
