摘要 目的检测ABCA1基因第7号外显子中R219KSNP的发生率,并探讨不同基因型与炎性因子IL-1βI、CAM-1水平的关系。方法冠心病患者112例及正常人108例,分别进行PCR反应及限制性酶切,确定不同的基因型,并检测每例血浆中炎性因子IL-1βI、CAM-1的水平。结果RR、RK、KK基因型之间进行多重比较。冠心病组IL-1β水平差异显著(P<0.01),对照组中IL-1β水平无明显差异(P>0.05);冠心病组及对照组ICAM-1水平差异显著(P<0.01)。以RR型最高,KK型最低。结论ABCA1基因第7号外显子中R219K位点K等位基因能显著降低ICAM-1的水平,能显著降低冠心病人的IL-1β水平,但对正常人IL-1β无明显影响。 Objective To detect the incidence of single nueleotide polymorphisms (SNP) of R219K in the seventh exon of ATP binding cassette transporter A1 gene (ABCA1) and the association between genotypes and the serum level of IL-1β and ICAM-1. Methods Genotypes were determined by PCR and restriction digestion techniques in 112 CAD cases and 108 control subjects. The levels of inflammatory cytokine IL-1β and ICAM-1 in patients plasma were determined. Results Comparisons among RR, RK and KK genotypes were made. The level of IL-1β in samples from the CAD group was variable(P〈0.01 ), while the level of IL-1β in samples from the control group was similar (P〉 0.05). The levels of IL-1β and ICAM-1 among RR, RK and KK genotypes were significantly different, with the highest level in patients with RR genotype and lowest in KK genotype. Conclusion The K allele of R219K SNP in the seventh exon of ABCA1 could remarkably decreases the level of ICAM-1 and IL-1β in CAD patients, but could not affect the level of IL-1β in healthy individual.
