大鼠脑损伤后星形胶质细胞结构和NO、ET的变化

Experimental Study on Changes of Astrocytes and NO and ET Level in Brain Tissue in Rats Following Traumatic Brain Injury

摘要 目的观察脑损伤后星形胶质细胞(Astrocyte,Ast)形态学变化及脑组织一氧化氮(n itric oxide,NO)、内皮素(endothlin,ET)含量变化。方法利用M arm arou创立的模型造成大鼠脑损伤,于伤后6h、24h、48h、120h胶质纤维酸性蛋白(glial fibrillary ac id ic prote in,GFAP)免疫组化染色观察Ast形态学变化并测脑组织NO、ET含量。结果脑损伤后6h出现GFAP表达的升高,48h达高峰,120h有回落但仍处于较高水平。脑组织NO和ET含量脑损伤后代偿性增加,24h-48h维持在较高水平,与对照组差异有统计学意义(P<0.01)。结论Ast、NO和ET在颅脑损伤的病理过程中有重要作用,且三者在损伤过程中存在一定变化规律。 Objective To investigate the changes of astrocytes (Ast) and NO and ET level in brain tissue in rats following traumatic brain injury. Methods Brain injury models were established by Marmarou model. The pathological changes of astrocytes after injury 6h,24h,48h and 120h were stud- ied by GFAP immunohistochemical staining and the changes of NO and ET in brain tissue of 40 rats were tested. Experimental rats were divided into groups according to the injury time. The results were compared with the normal animal. Results The expression of GFAP was increased at 6h, reached a peak at 48h,then decreased at 120h,but it was not back the normal level at 120h. NO and ET level in brain tissue were both increased after brain injury. NO level reached its peak at 48h while ET level at 24h( P 〈0. 01 ). Conclusion Regular expression of GFAP and changes of NO and ET and Ast level would appear after brain injury, they had taken part in the process of traumatic brain injury.
