摘要
利用RAPD分析构建了苎麻属植物21份木质素含量不同材料的指纹图谱,从150个随机引物筛选出清晰且多态性高的15个引物,共扩增出85条DNA片段,其中特异性带68条,占80%。同时对退火温度进行了优化,反应条件稳定,重复性好。采用UPGM A法对21个品种扩增的谱带进行遗传聚类分析,在D=0.0760处可分为三类8组,可说明供试品种的亲缘关系。
An optional reaction system suitable for the assay and usage of RAPD in ramie was established. The program with fine repoduciblity and high reliability was effectively used for RAPD analysis in ramie genetics and breeding. Genomic DNA of 21 ramie varieties of different lignin content were amplified by RAPD from 150 arbitrary 10 base primers. The results showed that all 85 bands were identi- fied. The polymorphic bands accounted for 80% of the total amplified bands. By drawing ploygenetic relationship through cluster of bands, the experimented accession could be divided among 3 types and 8 groups.
