摘要
探讨了利用正相硅胶-反相洗脱系统测定毛果芸香碱的HPLC色谱条件。结果表明,流动相(乙腈为主体)中的含水量、磷酸与三乙胺的用量、pH都程度不同地改变该药物的保留时间。最佳色谱条件是:检测波长217nm,流动相为含0.005%磷酸的80%乙腈(以1mol/LNaOH调节pH=5)。
The HPLC conditions ware studied for assaying pilocarpine by normalsilica column--reverse elution system. The retention time of piloarpine changed with theproportion of water to acetonitrile,amount of phosphoric acid and triethylamine and pHvalue of the mobile phase. At last the best chromatographic conditions had set. The detective wavelength is 217nm. The mobihe phase is 80% acetonitrile containing 0. 005% phosphoric acid and adjusted pH to 5 with 1mol/L sodium hydroxide.
