摘要
本文研究了由SV40早期启动子和增强子启动的β-半乳糖苷酶基因在HeLa细胞瞬时表达的动态情况.外源β-半乳糖苷酶基因经磷酸钙方法导入受体细胞50h后其酶活性为60milliunits,该酶活性维持一段时间后呈下降趋势,经转染270h后,该酶活性降为0.此外本文结果还表明,携带有β-半乳糖苷酶基因的质粒对受体细胞HeLa具有较高的转染效率.
In this study, tendecy of transient expression of lacZ gene driven by SV40 early promoter and enhancer was studied within Hela cells. Exogenous lacZ gene was introduced into recipient cells by using calcium phosphate-mediated transfection method. β-galactosidase activity is 60 milliunits at 50 hours after the transfection. From then on, β-galactosidase activity remains almost unchangable for a while, then decreased. After transfection for 270 hours, the activity is zero. In addition, transfection efficiency of the plasmid containing lacZ gene is high for Hela cells according to the assay of β-galactosidase activity.
