5-FC/CD∷UPRT联合基因对大鼠脑胶质瘤的治疗作用

摘要 目的探讨5-氟胞嘧啶(5-FC)/CD∷UPRT联合基因系统对大鼠脑胶质瘤的治疗作用。方法以逆转录病毒介导构建C6-CD∷UPRT细胞系,应用立体定向技术移植C6-CD∷UPRT细胞至SD大鼠尾状核,设立C6-pLXSN和C6细胞为对照组,建立脑胶质瘤荷瘤鼠模型,应用5-FC腹腔注射治疗,观察肿瘤大小、大鼠存活期、电镜观察细胞形态和流式细胞术检测肿瘤细胞的凋亡指标的变化。结果5-FC/CD∷UPRT治疗组(A组)与各对照组相比,A组肿瘤直径平均为(4.2±0.7)mm,远小于对照组的(8.3±1.3)mm,(P<0.01);对照组平均存活期(32±3)d,A组为(85±2)d(P<0.01);A组电镜显示核染色质分裂明显,并见典型的凋亡小体;A组肿瘤细胞中凋亡细胞所占百分率为19.36%。结论5-FC/CD∷UPRT联合基因对大鼠脑胶质瘤有显著的治疗效果。 Objective To investigate the therapeutic effects of 5-FC/CD ∷UPRT combinative gene on encephalic glioma xenograft in SD mice. Methods Reconstructed malignant glioma cells C6-CD∷ UPRT,C6-pLXSN and C6 were engrafted to the brain by third dimension direction finder. When tumors grew up for 3 weeks, mice were treated with 5-FC. The therapeutic effects were determined by measuring tumor size, survival period, and examing the tumor by electron microscope and FCM assays. Results Compared with control groups,the growth of tumors in test group was significantly suppressed. The average survival period of control group was about 32 days, which of test group extended to 85 days. The percentage of apoptotic cell was 19. 36% in the results of FCM assays. Conclusion 5-FC/CD∷UPRT combinative gene has a significant therapeutic effect on mice encephalic glioma.
