The Application of Differential Display of mRNA for Cloning Genes Related to Thyroid Carcinoma

摘要 目的:筛选并克隆甲状腺乳头状癌、甲状腺未分化癌与其正常甲状腺组织之间差异表达的基因。方法:应用mRNA差异显示技术克隆与鉴定出差异的cDNA片断,并对其进行NCBIGenBank数据库同源性检索。结果:成功亚克隆出8个基因片段,进行GenBank数据库同源性检索,这些基因可能与甲状腺癌的发生相关,尤其在甲状腺未分化癌及乳头状癌中均见与LMAN1基因同源的差异带,提示LMAN1基因很可能为甲状腺癌相关基因。结论:获得的差异表达片段为进一步克隆甲状腺癌相关基因及功能研究奠定了良好的基础。 Objective: To screen and clone the genes expressed differentially between thyroid carcinoma and the paraneoplastic thyroid tissue, including papillary carcinoma of the thyroid (PTC) and undifferentiated carcinoma of the thyroid (UTC). Methods: Differentially expressed eDNA fragments were cloned and identified using differential display of mRNA, and then a homology search of the NCBI GenBank database was conducted. Results: Eight eDNA fragments were successfully cloned and identified in NCBI GenBank. Those genes are possibly related to oncogenesis of thyroid carcinoma, especially the differentially expressed fragment homologous to LMAN1, which was detected in PTC and UTC. Conclusions: The differentially expressed fragments establish a favorable foundation for cloning genes related to thyroid carcinoma and continuing with their functional studies.
