摘要
目的探讨帕夫林(TGP)对骨质疏松模型大鼠骨密度、骨骼肌蛋白及相关细胞因子的影响。方法6月龄SD雌性大鼠行双侧卵巢切除术,建立骨质疏松动物模型。对照组行假手术。每组8只,共分4组。假手术组和卵巢切除组均用生理盐水灌胃,帕夫林低剂量+卵巢切除组、帕夫林高剂量+卵巢切除组分别给予TGP50mg/kg·d)、150mg/kg·d)灌胃。所有动物给药时间均为12周。处死大鼠后取血清测白细胞介素-6(IL-6)、肿瘤坏死因子(TNF-α)、胰岛素样生长因子-1(IGF-1)、转移生长因子-β(TGF-β),右侧股四头肌固定部位肌肉测定其蛋白总量及白蛋白含量,取右侧股骨及腰椎测骨密度。结果TGP组较卵巢切除组骨密度明显增高(P<0.05),TGP组骨骼肌总蛋白含量较卵巢切除组高(P<0.05),帕夫林低剂量组IL-6水平较卵巢切除组明显下降(P<0.05)。TGP高、低浓度与各检测指标间的相关性分析,无统计学差异(P>0.05)。结论TGP能够提高大鼠骨密度,增加骨骼肌蛋白含量,降低血清IL-6水平。
Objective To determine the influence of total glucosides of paeony (TGP) on bone density, protein level of skeletal muscle and relative cytokine of osteoporotic rats. Methods Six-month-old SD rats were shamed as control (8 rats) or were ovariectomized (24 rats). The ovariectomized (16 rats) +TGP low dose and high dose group were treated with 50 mg/(kg · d) TGP and 150 mg/(kg· d) TGP respectively for 12 weeks. After the animals were sacrificed, IL-6, TNF-α, IGF-1, TGF- β in serum, protein level of skeletal muscle in right quadriceps femoris, and bone density of right femur and lumbar vertebra were determined. Results Compared with the ovariectomized group, the bone density and the protein level of skeletal muscle in TGP group increased significantly from the baseline (P〈0.05). The level of IL-6 in TGP low dose group decreased significantly from the baseline (P〈0.05). Conclusion TGP can increase the bone density, improve protein level of skeletal muscle, and decrease the serum level of IL-6 of osteoporotic rats.
