α-突触核蛋白在正常大鼠脑神经元中的亚细胞定位 被引量:1

摘要 目的研究α_突触核蛋白(α_synuclein)在正常大鼠脑神经元中的亚细胞定位,为阐明其正常的生理功能提供线索。方法超小金标记结合银增强免疫电镜技术。结果α_突触核蛋白不仅存在于神经细胞的突触前终末,也见于神经细胞的轴突、胞浆与核中。在突触前终末,α_突触核蛋白的分布与突触小泡无明显的关系。在神经细胞核内,靠近核膜部分α_突触核蛋白的分布较为密集,细胞核中心部位α_突触核蛋白则较为稀疏。在胞浆和轴突中,α_突触核蛋白散在分布。另外,在线粒体中也有该蛋白表达。结论α_突触核蛋白广泛分布于神经元的各个部位,可能参与神经元的多种生理功能。 Objective To examine the subcellular localization of α-synuclein in rat neurons using EM. Methods Electron microscopy using ultra small immuno-gold detection and silver enhancement was used to study the subcellular localization of α-synuclein in the neurons of rat. Results In neurons, α-synuclein was present in the presynaptic terminals, axons, cytoplasm and nucleus. In the presynaptic terminals, it showed that α-synuclein was hardly associated with the synaptic vesicles. In the nucleus, there was a more dense distribution of α-synuclein inside the nuclear membrane, but a sparse distribution in the center, α-synuclein scattered in the axons and cytoplasm. In addition, α-synuclein was expressed in the mitochondria, which had not been reported before. Conclusion The results show that α-synuclein is spread widely and might have important role in physiological function of neurons.
