摘要 目的:探讨TGFβ1对Burkitt淋巴瘤细胞生长的抑制作用和对c myc基因及蛋白表达的影响。方法:于Burkitt淋巴瘤细胞株Jiyoye中加入5ng/mLTGFβ1做为实验组,不加TGFβ1做为对照组,分别培养24、48和72h,用MTT、RT PCR及Westernblot方法检测细胞生存率、c mycmRNA及蛋白的表达水平。结果:经TGFβ1处理24、48和72h的Jiyoye细胞生存率分别为(80.5±3.14)%、(70.5±3.77)%和(56.4±3.36)%,与对照组相比均显著降低(t=8.7880,P=0.0127;t=13.5709,P=0.0054;t=22.5006,P=0.0197)。经TGFβ1处理的Jiyoye细胞随时间的延长生存率降低,72h时的生存率与24h相比差异有统计学意义,t=8.1862,P=0.0012;c mycmRNA及蛋白的表达水平也明显变化,24h开始下降,48和72h时明显受抑制,与对照组相比差异有统计学意义,P=0.0000。结论:TGFβ1可呈时间依赖性抑制Jiyoye细胞的生长,其机制可能与抑制c myc基因及蛋白的表达有关。 OBJECTIVE: To investigate the inhibition of TGF-β1 on cell growth, e-myc gene and protein expression in Burkitt lymphoma cell line. METHODS: Five ng/mL TGF-β1 in Burkitt lymphoma cell line was added in the experimental group, and having no TGF-β1 in the same cell line in the control group. The two groups were cultured for 24, 48 and 72 h, respectively, and the cell survival rate, c-myc mRNA and protein expression were detected by using MTT, RT-PCR and Western Blotting analysis methods. RESULTS: The Jiyoye cell survival rates in the experimental group were (80. 5± 3. 14) 0±, (70. 5±3.77) % and (56.4±3. 360% at 24, 48 and 72 h, respectively. Compared with the control group, the cell survival rates were significantly lower in experimental group (t=8. 788 0,P=0. 012 7; t=13. 570 9,P=0. 005 4;t=22. 500 6,P=0. 019 7). The survival rate at 72 h was also significantly lower than that at 24 h in the experimental group (t=8. 186 2,P=0. 001 2). c-myc mRNA and protein expression began to reduce at 24 h, and in evidence inhibited at 48 h and 72 h, and this was significantly lower than that in the control group, P= 0. 000 0. CONCLUSIONS: The inhibition of TGF-β1 on Jiyoye cell gowth is dependent on the time process. The mechanism might be related with the inhibition of c-myc gene and protein expression.
