子宫内膜样腺癌组织中p53和p63基因的表达及其意义 被引量:2

摘要 目的:研究p53和p63基因在子宫内膜样腺癌(endometrial adenocarcinoma,EC)发生发展中的作用,及其与子宫内膜样腺癌临床病理特征的关系。方法:采用免疫组化SP法检测38例EC和23例子宫内膜增生过长(endometrial hy-perplasia,EH)及10例正常增生期子宫内膜(be-nign proliferative endometrium,BPE)组织中p53、p63基因的表达情况。结果:p53蛋白在EC中的阳性率为31·6%,明显高于EH组和BPE组,P值分别为0·0457和0·0389;p53的表达与EC的手术病理分期和肌层浸润深度有关,P值分别为2·19×10-4和1·436×10-6,而与组织学分级无关。p63蛋白在EC中的阳性率为81·6%,与EH组、BPE组比较,差异均有统计学意义,P值分别为9·58×10-7和5·59×10-5。p63的表达与EC的组织学分级、手术病理分期和肌层浸润深度均无关;p53和p63基因在EC中的表达呈正相关。结论:p53和p63均参与了EC的发生,二者可能起协同作用;p53更多地表达于高分期及深肌层浸润的EC组织中,p63在EC中呈高水平表达,说明p53与EC的浸润发展有关,p63在EC的发生中可能起癌基因的作用。 OBJECTIVE:To study the roles of p53 and p63 in tumorigenesis and development of endometrial adenocarcinoma (EC) and their correlation with clinicopathologic features. METHODS: The expressions of p53 and p63 were detected by the immunohistochemical stain in 38 cases of EC, 23 cases of endometrial hyperplasia (EH) and 10 cases of benign proliferative endometfium (BPE). RESULTS: The positive rate of p53 was 31.6% in EC, and the expression was significantly higher than those in EH and BPE, P = 0. 045 7 and 0. 038 9. The expression of p53 was associated with the stage and depth of myometrial invasion in EC (P=2. 19× 10^-4 and 1. 436× 10^-6), but was not associated with the histological grade. The positive rate of p63 was 81.6% in EC, there was a significant difference between EC and EH or BPE, P=9. 58×10^-7 and 5.59× 10^-5. The expression of 1363 was not associated with the histological grade, stage and depth of myometfial invasion in EC. There was a significantly positive correlation between p53 and p63. CONCLUSIONS: p53 and p63 are involved in carcinogenesis of EC, and there may be an assistant role between p53 and p63. The expression of p53 is more common in high grade and deep myometrial invasion carcinoma. The expression of p63 is highly expression in EC. These results suggest that p53 protein is related to the progression of malignant EC,and p63 may be an oncogene in tumorigenesis of EC.
