摘要 后基因组时代,蛋白质组学作为功能基因组学研究的核心技术之一,已成为生物医学领域强大的技术支撑,并为白内障疾病的研究提供了一个新的平台。白内障和老化被认为是一类构象性疾病。晶状体透明性与具有分子伴侣活性的α-晶体蛋白密切相关。白内障发生时,多种疾病特征性的蛋白质组改变,可导致α-晶体蛋白分子内或分子间的交联,分子伴侣活性下降,加速白内障的形成。就蛋白质组学在白内障发病机制研究中的应用进行综述。 Proteomics is one of the critical technologies in functional genomics in post-genome era. It has been the technological foundation in the medical field. Proteomics provides a new way to study cataract. Cataract and aging is believed as a kind of conformational diseases. The transparency is mainly related to the role of molecular chaperone in ~t-crystallin. Appling proteomics, we can detect various of characteristic proteome,which induce intramolecular or extramolecular aggregation in ~t-crystallin and reduce its activity and accelerate the cataract formation. Here application of proteomics in the cataract research is reviewed.
