3种脱钙液对牙体牙周联合切片H-E染色效果的对比 被引量:7

摘要 目的对不同脱钙液在牙体牙周联合组织H-E染色切片的效果进行比较,并初步探讨了乙二胺四乙酸二钠(EDTA)-微波技术在牙体牙周联合组织切片脱钙过程中的应用。方法取成年健康Beagle犬下颌骨牙体牙周联合组织做为标本,采用10%硝酸溶液、Krinstensen’s液、EDTA-微波辐射3种不同的脱钙方法进行脱钙,在完成组织脱钙后进行H-E染色,并在显微镜下观察组织结构及染色效果。结果EDTA-微波辐射完成组织脱钙约需53h,H-E染色效果良好,牙髓细胞、成牙本质细胞、牙周膜细胞和牙本质小管组织结构清晰;Krinstens-en’s液完成组织脱钙约需1500h,H-E染色效果较好,组织结构较清晰;10%硝酸溶液完成组织脱钙约需360h,H-E染色对比度差,组织结构模糊不清。结论应用EDTA-微波辐射技术能在相对较短的时间内完成牙体牙周联合组织的脱钙,并在H-E染色切片中较好地保存了牙体牙周组织的结构。 Objective To compare the effects of decalcification with different decalcifying solutions on the qualities of H-E staining of the teeth and periodontium combined specimens. Application of EDTA-Microwave in the decalcification process was also discussed. Methods Three different decalcifying solutions: 10% nitric acid, Krinstensen's solution and ethylenediamine tetra acetic acid disodium salt (EDTA-2Na) with microwave treatment were studied in the experiment. The teeth and periodontium combined specimens of mandible of healthy adult Beagle dog were chosen for experiments. Then sections were stained by hematoxylin-eosin(H-E). The structure of tissues and cells were surveyed with microscope. Results It took 53 hours to accomplish the decalcification by the method of EDTA-Microwave technique. The structures of specimens such as odontoblastic cells and periodontium cells were seen clearly. The specimens which were decalcified by Krinstensen's solution took about 1500 hours to decalcify completely. Anatomy of teeth and periodontium were less clear than that using EDTA-Microwave method. The specimensWhich were decalcified by 10% nitric acid took about 360 hours to decalcify, but the quality of H-E staining was poor. Conclusion The results with microwave treatment in EDTA solution for decalcification were best and the cost of time was least, and most structure of teeth and periodontium tissues still remained.
