摘要
采用豚鼠对大鼠异种心脏移植模型,应用化学发光法和比色法分别测定了移植心切取时、林格氏液(4℃)保存1小时和移植后1小时移植心整体超氧阴离子(O_2^-)发光峰值和心肌脂质过氧化物(MDA)含量,结合光镜和电镜检查,探讨移植心超急性排斥(超排)时组织损伤和氧自由基的关系。结果显示异种移植超排时可产生大量氧自由基,参与超排和多种组织损伤。前列腺素 E_1具有一定的抗自由基损伤作用,可稍延迟超排。
In order to determine the relationship between hyperacute xenograft rejection and oxygen free radical (OFR) formation,in the model of guinea pig to SD rat cardiac xenotrans- plantation guinea pig heart specimens were obtained at harvesting,after stored in Ringer's lactate solution (4℃) for 1 hour,and at 60 min after transplantation to assess histological changes and OFR production by light microscopy,electromicroscopy and luminol-enhanced chemiluminescence (LC).OFR formation increased significantlly in hyperacute rejection.These findings suggests that OFR may play important role in the pathogenesis of discordant xenograft rejection.
