摘要
本文采用水法浸提海带多糖,对海带多糖的最佳浸提及醇析条件进行了筛选,并对海带多糖进行了脱蛋白和脱色研究,以优化其提取条件。结果表明,海带多糖水法浸提的最佳浸提条件为:在40目的海带粉中加入海带重25倍的水于90℃浸提5h,最佳醇析条件为:将海带提取液浓缩5倍、添加4.5倍体积分数95%的乙醇,此时海带多糖的得率最高,为10.49%。常规Sevag试剂脱蛋白效果不理想,在海带多糖提取液中加入0.2%质量分数的木瓜蛋白酶于70℃下水解1h,能完全除去海带多糖中的蛋白质。H2O2氧化脱色较吸附脱色效果好,但用Al2O3层析柱(20mm×800mm),装柱600mm,脱色3次依然可以获得很好的脱色效果。
In this paper, the Laminaria Japonica Aresch polysaccharide was extracted with the mathod of hot water. In order to gain the optimum extracting condition , we had sieved the extracting and separating out by ethanol of Laminaria Japonica Aresch polysaccharide through deproteinization and decolorization. The results showed that the optimum extracting condition of Laminaria Japonica Aresch polysaccharide was 90℃ hot water as extractant, the ratio of material to extractant lg: 25mL, time 5 hr. The optimum separating out condition was the ratio of contentrated liquid to 95% (volume percent) ethanol 1:4. 5. And the yield of polysaccharide could be up to 10. 49%. The effect that the protein of Laminaria Japonica Aresch polysaccharide was been taken off by hydrolyzing by papain was better than the normal Sevag reagent's. We could gain Laminaria Japonica Aresch polysaccharide with ideal decoloring effect by Al2O3 column chromatography.
