摘要
植物甾醇是一种混合物,以往的定性方法有:化学特征反应鉴定法、薄层层析法、红外光谱法等,它们普遍存在的缺点是:鉴定能力低,对样品的纯度要求高,有的甚至安全性很差。因此,欲获得准确的结果,有效的鉴定手段是至关重要的。植物甾醇制品经氢氧化钾皂化后,用正己烷提取,非极性色谱柱分离,质谱检测器定性,附氢火焰检测器的气相色谱仪检测,内标法定量。试验结果表明:该方法线性范围宽,标准曲线(以豆甾醇计)的相关系数为0.9991,植物甾醇的平均回收率为98.66%99.51%,相对标准偏差为2.46%,且方法重现性好,准确度高,适用范围广。
Phytoserin is a kind of mixture. Its qualitative method included identification method of chemical characteristic reaction, thin-layer chromatography and infra - red spectrometry etc. Previously, they had universal disadvantages such as low identification ability, high demand on sample purity and even low security guarantee. Therefore, the effectual means to get accurate results are very vital. Phytoserin product is saponified by KOH, which is followed by being extracted by n - hexane and separated by non - ploar column. All the separated components are identified by mass spectra detector, detected on FID and phytoserin is quantitated by internal standard method. The results show that the method has broad linear scope and the correlation coefficient for phytoserin based on stigma sterol is 0. 9991. In addition, the mehod has good recovery 98. 66% - 99. 51%, low RSD 〈 2.46%, good reproducibility , high accuracy and wide scope of application.
