摘要
用原位杂交及免疫组化的方法检测11例硬化肝脏、7例正常肝脏中 C-H-ras、C-fos、C-jun 和 p^(53)基因的表达。结果,正常肝脏均为阴性;10例(91%)肝硬化中 C-H-ras 基因异常表达;5例检出 C-fos 基因的异常表达;1例检出 C-jun 基因表达;未检出突变型 P^(53)蛋白在硬化肝脏中的表达。提示,C-H—ras 和 C-fos 基因在肝硬化组织中的异常表达构成肝癌发生早期事件中基因异常的一部分。
Using in situ hybridization and immunohistochemistry,the expression of c-H-ras,c- fos,c-jun and p^(53)genes was studied in 11 human cirrhotic liver fragments and 7 histologically normal liver fragments.No overexpression of any oncogene mRNAs and of p^(53)protein was detected in histologi- cally normal livers.By contrast,10 out of 11 cirrhotic specimens(91%)displayed abnormally overex- pression of c-H-ras transcripts;five cirrhoytic samples revealed increased level of c-fos mRNA;only one case exhibited increased level of c-jun mRNA.No mutated p^(53)protein was found,as assessed by im- munohistochemistry,in the cirrhotic livers.These data suggest that the overexpression of ras and fos genes might constitute an early gene abnormality predisposing to the later development of hepatocellu- lar carcinoma.
