摘要
目的:研究芦荟多糖对体外培养人表皮细胞增殖活性及细胞内钙离子水平的影响。方法:在表皮细胞培养液中加入不同浓度芦荟多糖(25,50,100,200,400mg.L-1),应用细胞计数、四甲基偶氮唑蓝(MTT)法观察细胞增殖情况,采用Fura-2钙荧光指示剂测定细胞内钙离子浓度。结果:芦荟多糖作用2d后,100mg.L-1以上芦荟多糖组表皮细胞增殖明显增快(P<0.01),并呈剂量依赖性;与对照组比较,200mg.L-1和400mg.L-1芦荟多糖组表皮细胞内钙离子浓度显著性升高(P<0.05)。结论:芦荟多糖可能是通过调节细胞内钙离子浓度发挥其促进增殖作用。
OBJECTIVE To investigate the effects of polysaccharide (AP) from Aloe vera on proliferation and intracellular calcium levels of the in vitro cultured keratinocytes. METHODS The cultured keratinocytes were treated with AP at concentrations of 25,50,100, 200, 400 mg· L^- 1. The proliferation of keratinocytes was measured by cell count and MTT analysis, the concentration of intracellular calcium was determined using the fluorescent calcium indicator Fura-2. RESULTS Addition of AP led to a dose-dependent promotion of keratinocytes proliferation after 2 day of incubation, which was statistically significant for AP at concentrations over 100 mg·L^-1 (P〈0. 01 ). Compared with control group, the levels of intracellular calcium were significantly increased at 200 mg· L^-1 and 400 mg· L^-1 AP (P〈0. 05). CONCLUSION AP may regulate intracellular calcium which results in the proliferation of the keratinocytes.
