摘要
目的:研究杜仲改善糖尿病(DM)大鼠阴茎组织氧化损伤的效应,为开发新型治疗勃起功能障碍(ED)的药物提供实验依据。方法:随机选取的雄性DM大鼠20只和正常大鼠10只分为3组:A组(n=10,DM大鼠赋形剂灌胃组),B组(n=10,DM大鼠杜仲灌胃组)和C组(n=10,正常大鼠赋形剂灌胃对照组);灌胃4周后用透射电镜技术检查阴茎组织超微结构特征,并测定血清及阴茎组织匀浆液中超氧化物歧化酶(SOD)活性。结果:SOD活性:B组显著高于A组(P<0.01),C组显著高于A组(P<0.01),B组与C组比较差异无显著性。透射电镜观察:与C组大鼠比较,A组大鼠阴茎组织有髓神经纤维排布失序,部分变性、板层分离形成透明空泡或网络状,小血管、内皮细胞、平滑肌细胞和细胞器出现不同程度的损伤;B组大鼠阴茎组织有髓神经纤维髓鞘排布有序,内皮细胞、平滑肌细胞和细胞器未见明显异常。结论:杜仲可通过提高SOD活性改善DM大鼠阴茎组织氧化损伤。
OBJECTIVE To study the effects of eucommia ulmoides oliv on improving oxidation damage in penile tissue of diabetic rats,and provide experimental foundation for exploration of new drug treating erectile dysfunction. METHODS 20 male diabetic rats selected randomly and 10 normal rats were divided into three groups:group A (n = 10, diabetic rats were gavaged with escipient),group B (n = 10, diabetic rats were gavaged with eucornmia u lmoides oliv) and group C(n = 10, the normal rats were gavaged with escipient). After gavaged for four weeks, the ultrastructure of penile tissue was examined by transmission electron microscope,and superoxide dismutase(SOD) activitiies in serum and penile tissues were detected. RESULTS Compared with that in group A,SOD activities in group B and C were notably increased(P〈0. 01 ). There were no notable differiences between group B and C(P〉0. 05). The examination by transmission electron microscope showed that compared with group C, in penile tissues of the rats of group A, myelinated nerve fibers irregularly arranged and partially degenerated, myelin sheaths la mellas were splited and became vacuoles or network forms, and some certains of injuries were observed in small vessels, endothelial cells, smooth muscle cells and organell. While in group B, the myelin sheath of never fibers arranged in better orders and small vessels,endothelial cells,vascular smooth muscle cells and organell showed no obviouse abnormalities. CONCLUSION By enhancing SOD activity,eucornmia ulmoides oliv can improve oxidation damage in penile tissue of diabetic rats.
