益活清胰Ⅰ号对重症急性胰腺炎大鼠心肌组织中细胞凋亡调节蛋白表达的影响

Effects of Yihuoqingyi decoction Ⅰ on the intramyocardial expression of apoptosis-regulated protein in severe acute pancreatitis rats

摘要 [目的]研究益活清胰Ⅰ号对重症急性胰腺炎(SAP)大鼠心肌功能损害时细胞凋亡调节蛋白Bcl-2和Bax在心肌组织中表达的影响,并探讨其作用的机制。[方法]84只SD大鼠随机分为假手术组(正常组,n=12)、对照组(n=36)及治疗组(n=36)。SAP模型采用5%的牛磺胆酸钠胰胆管逆行注射方法建立。苏木精-伊红染色光镜下观察胰腺及心肌组织的病理变化,免疫组化法检测术后6、12、18 h心肌Bcl-2、Bax表达及核因子-κB(NF-κB)活化情况。[结果]治疗组胰腺及心肌组织的病理变化减轻,SAP各时间点均可见NF-κB明显活化,治疗组各时间点NF-κB活化与对照组相比均明显减弱(P<0.01)。术后6 h各组Bcl-2、Bax无显著性变化(P>0.05),12、18 h对照组Bcl-2的表达较正常组及治疗组显著减少(P<0.01),121、8 h对照组Bax的表达较正常组及治疗组显著增加(P<0.01)。[结论]益活清胰Ⅰ号可减轻NF-κB在心肌组织中的活化,减少SAP大鼠心肌组织Bax的表达及增强Bcl-2的表达,从而减轻心肌细胞凋亡,对SAP心肌组织具有保护作用。 [Objeetive] To investigate the effect of Yihuoqingyi decoction Ⅰ (YHQYD Ⅰ ) on the intramyocardial expression of apoptosis-regulated proteins in the severe acute pancreatitis (SAP) rats. [Methods] Eighty-four male SD rats were divided randomly into the sham-operated group (group A, n=12), the control group (group B, n=36) and the herbs treatment group (group C, n=36). SAP was induced by intraductal administration 5% sodium taurocholate. The expression of NF-kB, Bcl-2 and Bax in myocardial tissue was determined by SP immunohistochemical technique. Pancreatic and myocardial tissues were stained with HE and examined under light microscopy. [Results] Pathological changes in pancreatic and myocardial tissues of the treatment group alleviated in contrast to the control group. NF-kB activity in myocardial tissue increased significantly at 6h after induction of SAP, and myocardial NF-kB activity of the treatment group decreased significantly compared with that of the control group (P〈0. 01). Bel-2 and Bax expression in myocardial tissue had little changes at 6h after induction of SAP; Bel-2 expression of the control group in myocardial tissue decreased significantly, compared with that of the treatment group and the normal group (P〈0. 01) at 12, 18h after SAP; and Bax expression of the control group in myocardial tissue increased significantly compared with that of the treatment group and the normal group (P〈0.01) at 12, 18h after SAP. [Conclusion] YHQYD Ⅰ can regulate the expression of myocardial Bcl-2, Bax in the SAP rats, and alleviate myocardial apoptosis following SAP and have protective effect on the heart of SAP rats.
