摘要
目的观察丹参素对体外培养内皮祖细胞(EPC)数量和功能的影响。方法密度梯度离心法获取外周血单个核细胞,培养7 d后,收集贴壁细胞并加入丹参素,5、102、0 mg.L-1干预72 h。荧光显微镜和流式细胞仪鉴定EPC,分别观察EPC的数量、增殖、迁移及黏附能力。结果与对照组相比,丹参素干预组可促进外周血EPC扩增,并显著改善外周血EPC的黏附、迁移和增殖能力。结论丹参素可增加培养EPC的数量并改善其功能。
OBJECTIVE To investigate whether Danshensu can augment endothelial progenitor cell (EPC) number, and improve its function. METHODS Total mononuclear cells were isolated from peripheral blood by Ficoll density gradient centrifugation, and cultured in vitro. After culture for 7 days, attached cells were stimulated with Danshensu (5,10 and 20 mg·L^-1 ) for 72 h. The cells were identified by immunohistochemistry,flow cytometry and tested the abihty to intake the acLDL. EPC proliferation and migration were assayed by MTT assay and modified Boyden chamber assay. EPCs adhesion assay was performed by replating cells on fibronectin - coated dishes, and then counting adherent cells, RESULTS Danshensu could increase the number of EPC and promote EPC proliferative, migratory, adhesive capacity. CONCLUSION Danshensu can promote EPC augmentation and enhance its functional activity.
