摘要
目的探讨运动负荷对SD大鼠成骨细胞整合素(integrin)的表达和血清骨钙素(BGP)水平的影响。方法挑选♀3周龄SD大鼠,运用高电压、低电流动物刺激仪,刺激SD大鼠进行主动跑跳训练,成功地模拟出运动负荷的动物模型。取鼠后肢骨分离的成骨细胞,分别在含10%胎牛血清的F-12培养基中培养、传代,再采用流式细胞技术测定和分析整合素α2、β1、β3在成骨细胞膜上的表达量;取鼠心脏血采用液体闪烁仪检测成骨细胞BGP的表达。结果运动负荷可显著改变SD大鼠成骨细胞整合素的表达及血清骨钙素的水平。结论运动负荷可直接对大鼠骨的形成产生有益的影响。
OBJECTIVE To investigate that the effect of exercises on integrins and osteocalein in the osteoblasts of Sprague- Dawley (SD) rats. METHODS 3 - week - old female SD rats was chosed for high voltage and low current electrical stimulation, workload exercise model was successfully simttlatod.The osteoblasts isolated from SD rats femora were cultured in F - 12 medium containing with 10% fetal bovine serum and subcultured. The expression of integrins α2、β1、β3 on the membrane of osteoblasts were studied with Flow Cytometry (FCM) ; BGP in blood from hearts were detected with LSC. RESULTS Exercises improve serum BGP and expressions of integrins α2、β1、β3 in the femoral osteoblasts of SD rat' s significantly. CONCLUSION Workload exercises could be beneficial to achieve bone remodeling.
