摘要
目的 探讨醇溶性蟾蜍毒素在小鼠体内对艾氏腹水癌肿瘤细胞的抑制作用及其毒副作用。方法 本研究用乙醇溶解蟾蜍分泌物原浆,采用减压蒸馏等方法进一步部分分离纯化,获得醇溶性的蟾蜍毒素混合物(EET),将其作用于艾氏腹水癌荷瘤小鼠模型,计算小鼠的生命延长率,同时通过血生化和病理等检测观察其对各脏器的毒副作用。结果 5mg/M和0.5mg/M的EET对艾氏腹水癌荷瘤小鼠的生命延长率分别是42.6%(P〈0.05)和33.7%(P〈0.05);同时,0.5mg/M的EET可明显升高白细胞达(11.8±3.9)×10^9/L(P〈0.05)。但EET在5mg/M时可引起总胆红亲升高达(42.5±9.8)μmol/L(P〈0.05)。结论 0.5~5mg/M的EET可以明显抑制荷瘤小鼠体内艾氏腹水癌肿瘤细胞的生长,但EET的剂量达到5mg/kg时可以损伤肝脏。
Objective To study the inhibition effect of ethanol extract of toad venom (EET) on Ehrlich's ascites carcinoma and its toxicity in mice. Methods EET was purified by depressure distillation, then injected intraperitoneally or subcutaneously into the mice bearing Ehrlich's ascites carcinomas. The prolongation rate of life span in mice were calculated and the toxicity of EET was evaluated by biochemical parameters and pathological examination. Results The prolongation rate of life span in mice bearing Ehrlich's ascites carcinomas were 42.6% and 33.7% ,respectivdy (P〈 0.05). EET 0.5 mg/kg increased the number of white blood ceils to ( 11.8 ± 3.9) × 10^9/L ( P 〈 0.05) and EET 5 mg/kg increased the total bilirubin to (42.5 ± 9.8)μmol/L (P〈0.05). Conclsion These results suggest that EET 0.5 to 5 mg/kg could inhibit the growth of Ehrlich's ascites carcinoma in vivo in mice,but EET of 5 mg/kg may injure the liver.
