摘要
目的:探讨肿瘤坏死因子(TNF)-α与CVB3m重复感染导致的病毒性心肌炎的心室重构的关系。方法:连续4次注射CVB3m病毒液感染小鼠,分别于末次感染后5、10、306、0 d处死动物,Western Blot法测定心肌中基质金属蛋白酶(MMP)和组织型MMP抑制剂(TIMP)的蛋白含量;ELISA法检查血清中TNF-α水平;氯胺T氧化法检测心肌中胶原蛋白的含量。结果:心肌中的MMP-1和TIMP-1蛋白水平和血清中TNF-α变化趋势一致,即先升高,然后下降,但TIMP-1的变化稍延迟。心肌中的胶原蛋白水平则与MMP-1和TIMP-1蛋白含量比值呈负相关(r=-0.9903,P<0.01)。结论:CVB3m重复感染可导致血清中TNF-α水平持续升高,增加的TNF-α与MMP-1和TIMP-1的高水平表达有关,TNF-α与病毒重复感染导致的心室重构关系密切。
Objective:To investigate the relationship between tumor necrosis factor-α (TNF-α)and ventricular remodeling in myocarditis caused by repetitive CVB3m infection. Method:Four week old mice were infected with CVB3m 4 times successively, killed at 5,10,30,60 days later separately after the final infection. Western Blot for determining the content of MMP-1 and TIMP-1, Chloramine T oxidation for gauging content of collagen in myocardium, ELISA for measuring TNF-α level in serum. Result: It showed there was the trend of TNF-α was consistent, MMP-1 and TIMP-1 with, however TIMP-1 changed later more slowly. The collagen in myocardium was positively correlated with protein ratio of MMP1 and TIMP-1. Conclusion:Repetitive CVB3m infection can contin uously increase TNF-α in serum There is a positive correlation between high TNF-α level and expression of MMP-1 and TIMP-1 . So TNF-α is closely correlated with ventricular remodeling caused by repetitive CVB3m infection.
