吡喹酮与干扰素-γ对血吸虫病小鼠肝窦病变的影响 被引量:2

摘要 目的:观察吡喹酮和干扰素-γ(IFN-γ)对血吸虫病小鼠肝窦病变的影响。方法:将感染日本血吸虫小鼠在16周时随机分成三组:吡喹酮治疗组、吡喹酮联合IFN-γ治疗组以及模型组,治疗8周,另取10只正常小鼠为对照组。肝标本行常规病理染色,进行肝纤维化程度的半定量分析,免疫组化染色检测肝窦IV型胶原(C-IV)和第Ⅷ因子相关抗原(vWF)的表达,另每组取2例小鼠肝组织进行肝窦透射电镜的检查。结果:吡喹酮治疗组肝窦内皮细胞(SEC)未见窗孔,可见基底膜(BM),肝纤维化程度、肝窦C-IV和vWF的表达下降均不明显(P>0.05)。吡喹酮联合IFN-γ治疗组SEC可见少数窗孔,未见BM,肝纤维化程度、肝窦C-IV和vWF的表达均下降明显(P<0.05或P<0.01)。结论:IFN-γ可改善血吸虫病肝窦病变的作用,并可能据此作用实现其抗肝纤维化作用。 Objective:To observe the effect of praziquanteland and interferon-γ on liver sinusoid of mice infected by schistosoma japonicum. Methods:The mice were divided randomly into three groups after infection at 16 week:infection group, praziquantel treated alone group and the combination of praziquantel and interferon-γ treated group. The infected mice were treated for 8 weeks and 10 normal mice were as the control. The liver specimens of mice were used for pathological examination to get to know the degree of liver fibrois by semi-quantify and observe the expression of C-Ⅳ and vWF in liver sinusoid by immuhistochemisty staining. The ultramicrostructure of the liver tissue was observed by transmission electron microscope.Results:Mter chemotherapy with praziquantel, the fenestration in SEC could not be observed, the base membrane could be observed, the liver fibrosis degree, the expression of C-Ⅳ and vWF in liver sinusoid remained consisitent with infection group (P〉0.05). When treated with praziquantel and interferon-γ the fenestration in SEC could be observed, but the base membrane could not be observed, the liver fibrosis degree, the expression of C-Ⅳ and vWF in liver sinusoid decreased obviously (P〈0.05 or P〈0.01). Conclusion:Interferon-γ can reverse pathological changes in liver sinusoid induced by sehistosoma japoncium, and it's anti-fibrotie effect may depend on this.
