摘要
目的:观察蝎毒(scorp ion venom,SV)及其分离组分对4种人肿瘤细胞(肺癌A549细胞、人低分化胃腺癌细胞BGC-823、人肝癌细胞SMMC-7721、人骨髓细胞白血病细胞HL-60)生长的抑制作用。方法:采用Sephadex G-50柱层析法分离蝎毒;用MTT法检测肿瘤细胞的存活率。分别以丝裂霉素C(MMC)及顺铂(C-DDP)为阳性对照药,以蝎毒及其组分1 mg/L、10 mg/L和100 mg/L的浓度处理4种人肿瘤细胞,作用48 h后,分别观察蝎毒及其分离各组分对上述细胞的影响。结果:用Sephadex G-50柱层析法分得6个峰(APⅠ1,API2,APⅡ,APⅢ1,APⅢ2,APⅣ),其中蝎毒组分APⅢ1(antineop lastic polypeptideⅢ1)对HL-60,SMMC-7721,BGC和A549细胞生长有明显的抑制作用,与对照组比较差异有显著性(P<0.05,或P<0.01)。对A549细胞和HL-60细胞具有较好的剂量-效应关系。结论:蝎毒分离6个组分中,蝎毒组分APⅢ1对HL-60,SMMC-7721,BGC和A549 4种人肿瘤细胞具有明显的生长抑制作用,剂量效应关系明显。
Objective:To observe the inhibitory effects of scorpion venom from Buthus Martensii Karsch and its components on human tumor cell lines (HL-60, SMMC-7721, BGC and A549). Methods: The components were isolated from scorpion venom by Sephadex G-50 gel column chromatography. MTT method was used to detect the survival of tumor cells (HL-60, SMMC-7721, BGC and A549) after treated with SV and its components in vitro at different final concentrations (1 mg/L, 10 mg/L and 100 mg/L), compared with outcomes of Mitomycin C (MMC) and cis-platinum (C-DDP) treatments as positive controls. The effects of SV and its components on tumor cells mentioned above were observed at 48 hours after exposure. Results: Sephadex G-50 gel column chromatography identified6 peaks (APⅠ1,APⅠ2,APⅡ,APⅢ1,APⅢ2,APⅣ), of which, AP Ⅲ4 showed obvious cytotoxicity on tumor cells of HL-60, SMMC-7721, BGC and A549 (P 〈 0.05, or P 〈 0.01 ) , and obvious dose- response effects on HL-60 and A549. Conclusion: AP Ⅲ1 distinctly inhibited the growth of HL-60, SMMC-7721, BGC and A549 cell lines.
