摘要
目的观察静脉麻醉药异丙酚对N甲基D天冬氨酸(NMDA)致PC12细胞损伤时氨基酸水平的影响,并探讨异丙酚细胞保护作用的可能机制。方法建立300μmol/LNMDA诱导的PC12细胞损伤模型,采用高效液相色谱分析法测定PC12细胞氨基酸含量。结果300μmol/LNMDA处理4h可使PC12细胞谷氨酸含量明显增加,但对天冬氨酸、谷氨酰胺、甘氨酸含量无明显影响;12.5和125μmol/L异丙酚与300μmol/LNMDA同时处理PC12细胞4h后,谷氨酸含量较单纯NMDA组明显降低(P<0.05)。结论异丙酚可明显抑制NMDA诱导PC12细胞损伤所致的谷氨酸合成或释放,提示异丙酚可能通过抑制谷氨酸的合成或释放而产生细胞保护作用。
Objective To observe the influence of propofol on amino acid levels in cultured PC12 cells impaired by N-methyl-D-aspartare, and explore the possible protective action mechanism of anesthetic propofol. Methods The levels of amino acid were measured by high performance liquid chromatography (HPLC). Results After exposing to NMDA 300μmol/L for 4h, the release of glutamate levels from PC12 cells was increased significantly, whereas the release of glutamine, aspartate, and glysine levels remained unchanged. In the presence of propofol 12. 5, 125μmol/L for 4h, the levels of glutamate decreased significantly (P〈0. 05, compared with NMDA-treated group). Conclusion Propofol can inhibit the NMDA-induced release of glutamate from PC12 cells, suggesting that the cytoprotection effect of propofol may be related to its inhibitory action on the release of glutamate.
