摘要
目的探讨染色体微卫星不稳定性和杂合性缺失与急性白血病发生的相关性。方法应用PCR-非变性聚丙烯酰胺凝胶电泳-银染技术检测7q22-36微卫星不稳定性和杂合性缺失现象。结果在24例急性粒细胞性白血病中,有9例发生了杂合性缺失,发生率为37·50%,D7S515、D7S523、D7S636、D7S487的发生率分别为16·67%(4/24)、8·33%(2/24)、8·33%(2/24)和4·17%(1/24)。结论白血病患者染色体上的多位点微卫星改变,提示白血病的发生、发展与染色体的多部位、多步骤改变有关。
Objective To explore the frequency of loss of heterozygosity (LOH) on the long arm of chromosome 7 and the relationship between LOH and acute leukemia. Methods PCR-polyacrylamide gel electrophoresis-silver staining technique was used to test the LOH of Microsatellite DNA sites located in 7q22-36. Results The total frequency of LOH in 24 patients with AML was 37.5%. The frequency of LOH at D7S515, D7S523, D7S636 and D7S487 was 16.67%, 8.33%, 8.33% and 4.17% respectively. Conclusion Microsatellite alterations of multilocus observed in leukemia indicate that the pathogenesis and progress of leukemia is related to multi-region and multi-step changes of chromosomes.
