摘要 目的探讨全氟辛烷磺酰基化合物(PFOS)对脑血管内皮细胞的损伤作用。方法以不同浓度的PFOS处理脑血管内皮细胞(HBMEC),30min后观察细胞形态;用免疫荧光染色的方法观察细胞骨架微管蛋白α-tubulin的表达;不同剂量PFOS连续腹腔内注射14d观察大鼠行为学改变。结果随着PFOS浓度的增高细胞形态发生变化,细胞变圆、部分细胞不再贴壁、细胞骨架微管解聚细胞形态变圆,微管逐渐向细胞核周围聚集;动物实验表明高浓度PFOS可导致大鼠脑损伤、出现偏瘫的体征。结论全氟辛烷磺酰基化合物(PFOS)对脑血管内皮细胞有损伤作用。 [Objective] To evaluate the damaging effects of perfluorooctanesulfonate (PFOS) on brain tissue. [Methods] Human brain microvascular endothelia cells (HBMEC) were treated with PFOS at different concentration in vitro and the morphologic change of HBMEC was observed after 30 minutes, immunofluorescence method as used to locate cell skeleton protein α-tubulin change of HBMEC; we set up animal models and PFOS at different dosages was injected 14 days continuously to observe the behavior change of the rats. [Results] Morphologic change of HBMEC is that cells became round and some cells floated above the wall with PFOS stressing; cell skeleton protein α-tubulin unbinded and assembled around cell nucleus; animal model showed PFOS at high dosages could damage brain tissue of rats. [Conclusion] PFOS at high dosages could damage brain tissue.
