保元排毒丸对顺铂致LLC-PK1细胞毒性的保护作用 被引量:1

摘要 目的:观察保元排毒丸对顺铂所致猪肾小管上皮细胞(LLC-PK1)损伤的影响及超氧化物岐化酶(SOD)和一氧化氮(NO)的变化。方法:LLC-PK1细胞分4组培养,空白血清组采用不含药血清进行培养,含药血清组以保元排毒丸含药血清进行培养,模型组以不同浓度的顺铂进行培养,模型+含药血清组是在不同浓度的顺铂基础上予以10%保元排毒丸含药血清进行培养。在培养12h和24h时以MTT法测定OD值,计算细胞存活率。培养24h时,取细胞液测定SOD和NO含量。结果:与空白血清组相比含药血清组OD值明显升高(P<0.01),模型+含药血清组细胞存活率明显高于模型组(P<0.01),模型+含药血清组细胞液NO含量明显低于模型组(P<0.01)。结论:保元排毒丸能促进LLC-PK1细胞的增殖,并具有抗氧化功能,能预防或降低顺铂引起的肾毒性。 Objective:To investigate effect of Baoyuan Paidu Wan on injuries of pig LLC- PK1 ells induced by cisplatin and the changes of SOD and NO value. Methods: LLC- PK1 cells were divided into four groups for culture. Cells were respectively cultivated in blank serum group, in serum with Baoyuan Paidu Wan group, in model group with cisplatin of different concentration and in model group with cisplatin of different concentration plus 10% serum with Baoyuan Paidu Wan. The OD value was measured with MTT method in 12 h and 24 h after the culture. Then the cells livability was calculated according to MTT results. The content of SOD and NO were tested in the cell fluid in 24 h after the culture. Results: Compared to blank serum group, the OD value was increased significantly in serum with Baoyuan Paidu Wan group( P 〈 0.01 ). The cell livability in model plus serum with Baoyuan Paidu Wan group was obviously higher titan that in model group (P〈 0.01 ). The content of NO in cell fluid in model plus serum with Baoyuan Paidu Wan group was significantly lower than that in the model group( P 〈 0.01 ). Conclusion: Baoyuan Paidu Wan could promote the proliferation of LLC- PK1 dis and decrease nephrotoxicity caused by cisplatin, which might be through its anti - oxidation function.
