摘要
为了得到纯度较高的磷脂酰胆碱,对乙醇冷冻提取法纯化磷脂酰胆碱的工艺进行了研究。采用高效液相色谱法测定磷脂酰胆碱的含量,通过正交实验设计考察了乙醇浓度、料液比和提取时间等因素在不同水平下对磷脂酰胆碱含量的影响,对试验结果进行了方差分析和显著性检验。结果显示乙醇冷冻提取法最佳工艺组合为A2B3C3,即93%乙醇溶液,料液比1:3,冷冻条件下提取8.5h,在选定的最佳工艺条件下得到的磷脂酰胆碱的含量由原来的33%提高到了75%,可见乙醇冷冻提取法能有效地纯化磷脂酰胆碱。
The purification of phosphotidylcholine (PC) with alcohol solution in frozen condition was studied. The influence of alcohol content, the ration of material to solvent and extracting time in different levels was performed by orthogonal plan and content of PC was determined by HPLC. The results showed that the optimal condition for the purification of PC with alcohol solution in frozen condition was 1:3 of 93 % alcohol solution to the sample weight ratio, and 8.5 h, and content of the obtained PC increased up to 75 %, which indicated that high content of PC could be achieved by the method of extracting with alcohol solution in frozen condition.
