摘要
目的探讨大蒜素对小鼠膀胱癌的抗肿瘤效果和机制。方法将100μg/ml大蒜素直接加入肿瘤细胞培养6h,用相差显微镜观察肿瘤细胞形态学变化。MTT法测定大蒜素对肿瘤细胞体外生长的直接影响。动物实验检测大蒜素的抗癌效果。在实验第19天切除肿瘤组织作病理学检查,了解免疫反应情况。LDH释放法测大蒜素注射后淋巴细胞对肿瘤细胞的细胞毒活性。结果大蒜素对膀胱肿瘤细胞有明显的直接细胞毒作用,与生理盐水对照组相比,大蒜素剂量为0.1mg/ml时,肿瘤的抑制率可达56%,剂量为2.5mg/ml时,肿瘤抑制率可达99%。LDH释放法测得大蒜素注射后,能产生针对肿瘤细胞的细胞毒淋巴细胞。结论大蒜素对膀胱肿瘤有明显的抑制作用。
Objective To understand anti-tumor effect and mechanism of allicin for murine bladder tumor. Methods 100μg/ml allicin was added to the tumor cell culture for 6h,the morphology of tumor cells was observed by discrepancy microscope. The direct effects of allicin in vitro was evaluated in the MTT assay. Anti-tumor effect of aUicin in vivo was determined by animal experiment. On day 19 the tumors were excised and examined macroscopi- cally for evidence of immune response. CTL activity of lymphocyte was measured by the release of endogenous lactate dehydrogenase(LDH). Results Our experience with the observing morphology of MBT-2 cells in vitro and MTT assay suggested that allicin had significant direct cytotoxicity to bladder cancer ceils. In high dose group, it showed a marked delay in the appearance and growth of tumors after s. c. injection compared with control group(P 〈 0.01 ). Histology studies suggested that there were more macrophages, lymphocytes and fibroblasts at the tumor margin than control group. Immuned with allicin generated the cytotoxic lymphocyte responses to MBT-2 tumor cells. Conclusion Allicin has a marked anti-tumor effect on bladder tumor. This effect may be related to direct cytotoxicity and immune response. It may be an effective intravesical treatment agent for superficial bladder cancer.
