摘要
目的:建立一种新的栀子中京尼平苷含量测定方法。方法:研究了京尼平苷被β-葡萄糖苷酶水解的最佳条件及其水解产物京尼平与氨基酸的显色反应条件,水解温度为50℃,水解 pH 为5.0,水解时间为40min,京尼平苷液与酶溶液的体积比为1:1,水解完全后按体积比1:1加入精氨酸(Arg)溶液,80℃下显色1h,检测波长590nm。结果:该方法检测线性范围为1.3~52.8μg·mL^(-1),相关系数为0.9998,检测限达到0.66μg·mL^(-1),精密度为1.2%,回收率为99.8%~101.0%。结论:本方法操作简便,结果准确可靠。
Objective:The spectrophotometric method was established to determinate the geniposide from Gardenia. Method : The hydrolyze conditions of geniposide with β - glucosidase and the color reaction conditions were studied in this paper. The optimum conditions were : hydrolyze temperature at 50 ℃ for 40 min, hydrolyze pH 5.0, the volume ratio of geniposide solvent to β - glucosidase solvent 1 : 1, color reaction temperature 80 ℃, color reaction time 1 h, the volume ratio of geniposide solvent to Arg solvent 1 : 1. Result : The measurement range was 1.3 μg·mL^-1 to 52. 8 μg·mL^-1,the linear correlation was 0. 9998 ,the detection limit was 0. 66μg·mL^-1 ,RSD was 1.2% ,the recovery rate was 99. 8% to 101.0%. Conclusion:This method has the advantages of wide measurement range and low detection limit and it's simple and convenient to be used for the content determination of geniposide from gardenia.
