加速溶剂萃取法用于灯盏细辛中灯盏乙素的测定 被引量:14

摘要 目的:筛选加速溶剂萃取(ASE)的最佳提取条件,并比较其与常规提取方法的优劣。方法:以灯盏花药材为模型药物,运用正交试验法,色谱柱:Diamonsil ODS 柱(4.6mm×250mm,5μm),以乙腈和0.5%磷酸溶液进行 HPLC 梯度洗脱,检测波长335nm,流速:1.0mL·min^(-1),测定样品中灯盏乙素的含量。结果:ASE 提取灯盏乙素的最佳条件为:总静态提取时间:20min;提取温度:120℃;循环次数:3次;冲洗溶剂量:80%。结论:ASE 提取较常用回流提取法方便、快捷、效率高。 Objectives:To select the best conditions of ASE and compare it with regular extractive. Method:As the model of Erigern breviscapus ( Vant. ) Hant. - Mazz. , to determinate the content of scutellarein -7 - 0 - glucuronide with HPLC by gradient elution of orthogonal design. The column was Diamonsil ODS (250 mm ×4. 6 mm,5 μm) acetonitrile and 0.05 mol·L^-1 phosphoric acid solution were used as the mobile phase. The detection wavelength was 335 nm and flow rate was 1.0 mL·min^-1. Result : The best conditions of ASE were as follow : total static time : 20 min ;extractive temperature: 120 ℃ ;cycle :3 times ;flush volume :80%. Conclusion:The ASE has the advantages of convenient, fast and higher efficiency.
